Glycosylation Analysis of Human Serum Transferrin Glycoforms Using Pellicular Anion-Exchange Chromatography

چکیده
INTRODUCTION Glycoforms and Protein Sialylation Glycoproteins are proteins with a carbohydrate attached to the polypeptide backbones through one or more glycosylation sites. Oligosaccharides can be linked to a protein through a serine or a threonine as O-linked glycans, or through an asparagine as N-linked glycans. Glycoprotein glycoforms contain identical polypeptide backbones and differ from one another in the oligosaccharides attached to the glycosylation sites. To complicate the analytical task, the oligosaccharides attached to glycoproteins can also be sialylated at the nonreducing end of the glycans and the degree of sialylation can vary. Thus, the variation in glycoprotein sialylation can also create a collection of glycoforms.

متن کاملImproved HPLC method for carbohydrate-deficient transferrin in serum.
BACKGROUND There is need for a reference method for transferrin glycoforms in serum to which routine immunologic methods for the alcohol marker carbohydrate-deficient transferrin (CDT) can be traceable. We describe an improved HPLC method for transferrin glycoforms. METHODS Transferrin was iron-saturated by mixing the serum with ferric nitrilotriacetic acid, and lipoproteins were precipitated...
متن کاملDisialo-trisialo bridging of transferrin is due to increased branching and fucosylation of the carbohydrate moiety.
BACKGROUND Carbohydrate deficient transferrin (CDT) is used for detection of alcohol abuse and follow-up. High performance liquid chromatography (HPLC) of transferrin glycoforms is highly specific for identification of alcohol abuse, but unresolved disialo- and trisialotransferrin glycoforms sometimes makes interpretation difficult. The cause of this phenomenon is unknown, cannot be explained b...
متن کاملSeparation of transferrin types in human plasma by anion-exchange high-performance liquid chromatography.
An anion-exchange high-performance liquid chromatographic procedure was developed for the separation of transferrin in human plasma. The procedure allowed the four molecular forms of transferrin, which differ with respect to bound iron, to be separated from each other and from other plasma proteins. Transferrin variants, including B and D types, could also be identified using anion-exchange hig...
متن کاملChanges in Glycosylation of Alpha-1-Protease Inhibitor in Inflammation (Rheumatoid Arthritis and Crohn\'s Disease)
Alpha-1-proteinase inhibitor (API) is one of the acute phase proteins. Following an inflammatory stimuli the concentration of API increased up to four folds. Accompanying these quantitative changes, there is qualitative alterations in the structure of carbohydrate moiety (glycosylation). To determine the alterations in the glycosylation of API in inflammation, API was isolated from the sera of...
